The present invention relates to biscarbodiimides, thiourea derivatives, urea derivatives, and cross-linked hyaluronan derivatives having at least one intramolecular disulfide bond, and methods of preparation thereof. The invention also includes thiolated hyaluronan derivatives and salts thereof having at least one pendant thiol group or a modified pendant thiol group, and methods of preparation thereof. An example of a modified pendant thiol group is a sulfhydryl group linked to a small molecule such as a bioactive agent, for example a drug or pharmaceutically active moiety. A hyaluronan derivative having a sulfhydryl group linked to a pharmaceutically active moiety is useful as a sustained or controlled release drug delivery vehicle. Compositions containing the hyaluronan derivatives of the invention can reversibly viscosify in vivo or in vitro, in response to mild changes in condition, and are thus useful in ophthalmic surgery and in tissue engineering.
La présente invention concerne des biscarbodiimides, des dérivés thiourée, des dérivés urée et des dérivés hyaluronane réticulés présentant au moins une liaison bisulfure intramoléculaire, et les procédés de préparation correspondant. L'invention concerne également des dérivés hyaluronane thiolés et certains de leurs sels portant au moins un groupe thiol pendant ou un groupe thiol pendant modifié, et les procédés de préparation correspondants. Un tel groupe thiol pendant modifié est principalement un groupe sulfhydryle lié à une petite molécule telles qu'un agent bioactif, notamment un médicament ou un groupe fonctionnel pharmaceutiquement actif. Un tel dérivé hyaluronane portant un groupe sulfhydryle lié à un groupe fonctionnel pharmaceutiquement actif convient comme vecteur d'administration de médicament à libération prolongée ou contrôlée. Les compositions contenant les dérivés hyaluronane de l'invention sont capables de devenir réversiblement visqueuses in vivo ou in vitro en réaction à de légères modifications des conditions, ce qui fait qu'elles conviennent particulièrement en chirurgie ophtalmique et en génie tissulaire.
