A process for preparing thrombin which comprises treating a mixture comprising prothrombin, Factor Xa, Factor Va and phospholipids with calcium ions, at a pH less than pH 7.0 is provided. In particular the pH of less than pH 7.0 may be generated by the addition of the calcium ions or by buffering the preparation to less than pH 7Ø Thrombin preparations so produced may be subjected to further purification and are particularly stable even when substantially free of exogenous stabilising agents such as proteins, sugars, polyol and mixtures thereof, and may be subjected to freeze-drying and a virus inactivation by heat-treatment.

Le procédé faisant l'objet de cette invention, qui sert à préparer de la thrombine, consiste à traiter un mélange comprenant de la prothrombine, le facteur Xa, le facteur Va et des phospholipides avec des ions de calcium, à un pH inférieur à 7,0. Plus particulièrement, le pH inférieur à 7,0 peut être produit par l'addition des ions de calcium ou par tamponnage de cette préparation à un pH inférieur à 7,0. Les préparations de thrombine ainsi obtenues peuvent être soumises à une purification ultérieure et elles sont particulièrement stables même lorsqu'elles sont pour ainsi dire exemptes d'agents de stabilisation exogène, tels que protéines, sucres, polyol et mélanges de ceux-ci, et elles peuvent être soumises à une lyophilisation et à une inactivation virale par traitement thermique.
