It is intended to provide a TLR ligand/IL-1 response-injured nonhuman animal model in which the IL-6 production responding to TLR ligand/IL-1 is injured; and a method of screening a promoter or an inhibitor of the TLR ligand/IL-1 response or a preventive/remedy for atopic dermatitis-like inflammatory skin disease lesions by using this nonhuman animal. A TLR ligand/IL-1 nonresponsive nonhuman animal model, in which an endogenous gene encoding I.kappa.B-.zeta. of a nonhuman animal has been entirely or partly inactivated by a gene mutation such as breakage, deletion or substitution so as to lose the function of expressing I.kappa.B-.zeta. and thus the IL-6 production responding to the TLR ligand and IL-1 is injured, is constructed. By using this nonhuman animal and a test substance, a promoter or an inhibitor of the TLR ligand/IL-1 response or a preventive/remedy for atopic dermatitis-like inflammatory skin disease lesions is screened.

L'intention est de fournir un modèle d'animal non humain ayant une réponse à un ligand de TLR/à l'IL-1 abîmée dans lequel la production d'IL-6 répondant au ligand de TLR/à l'IL-1 est abîmée ; et un procédé de recherche par criblage d'un promoteur ou d'un inhibiteur de la réponse à un ligand de TLR/à l'IL-1 ou d'un agent de prévention/remède pour des lésions accompagnant des maladies inflammatoires de la peau comme la dermite atopique en utilisant cet animal non humain. On construit un modèle d'animal non humain ne répondant pas à un ligand de TLR/à l'IL1, dans lequel un gène endogène codant pour la I.kappa.B-.zeta. d'un animal non humain a été entièrement ou partiellement inactivé par une mutation génétique telle qu'une cassure, une délétion ou une substitution de façon à perdre la fonction d'expression de I.kappa.B-.zeta. et ainsi la production d'IL-6 répondant au ligand de TLR et à l'IL-1 est abîmée. En utilisant cet animal non humain et une substance à tester, on recherche par criblage un promoteur ou un inhibiteur de la réponse à un ligand de TLR/à l'IL-1 ou un agent de prévention/remède pour des lésions accompagnant des maladies inflammatoires de la peau comme la dermite atopique.
