A metalloprotease that converts TNF-.alpha. from the 26kD cell form to the cell form to the 17 kD form has been isolated and purified and the cDNA sequence known. In particular, the protease has a molecular weight of approximately 80 kD. The isolated and purified protease is useful for designing an inhibitor thereof, and may find use as a therapeutic agent. Assays for detecting the protease-inhibiting activity of a molecule are also an aspect of the invention.
Une métalloprotéase convertissant le facteur de nécrose tumorale alpha (TNF- alpha ) de la forme cellulaire de 26 kD en la forme cellulaire 17 kD a été isolée et purifiée et sa séquence d'ADN complémentaire, rendue connue. La protéase présente notamment un poids moléculaire d'environ 80 kD. Cette protéase isolée et purifiée se révèle utile pour la conception de son inhibiteur, de même qu'elle peut être employée comme agent thérapeutique. Des techniques de détection de l'activité inhibitrice de la protéase d'une molécule constituent également un aspect de cette invention.
