A controlled release transdermal system for the delivery of at least one therapeutic agent comprises: a reservoir comprising (a) the therapeutic agent in ionized form and (b) a pH adjusting agent which upon uptake of water is converted to a buffer solution and (c) a cyclized polysaccharide selected from a group consisting of cyclodextrin, cyclodextrin derivative and cyclodextrin polymer, the cyclized polysaccharide being capable of improving the solubility of the therapeutic agent in the buffer solution by forming an inclusion complex with the therapeutic agent; and a reservoir wall comprising a polymer being substantially impermeable to the ionized form or to the inclusion complex form of the therapeutic agent but permeable to water and to the unionized form of the therapeutic agent. The system allows the release of a therapeutic agent which may be a weak acid or base in a manner which shows less variation between patients than previous systems.
Un système transdermique à libération régulée pour l'administration d'au moins un agent thérapeutique comprend un réservoir contenant (a) l'agent thérapeutique sous forme ionisée, (b) un agent de régulation du pH qui, lors de la prise d'eau, se convertit en une solution tampon et (c) un polysaccharide cyclisé sélectionné dans un groupe comprenant la cyclodextrine, un dérivé de cyclodextrine et un polymère de cyclodextrine, le polysaccharide cyclisé pouvant améliorer la solubilité de l'agent thérapeutique dans la solution tampon en formant un complexe d'inclusion avec l'agent thérapeutique; et une paroi du réservoir comprenant un polymère sensiblement imperméable à la forme ionisée ou à la forme complexe d'inclusion de l'agent thérapeutique mais perméable à l'eau et à la forme non ionisée de l'agent thérapeutique. Le système permet la libération d'un agent thérapeutique qui peut être un acide faible ou une base et d'une manière telle qu'il y ait moins de variations d'un patient à un autre qu'avec les systèmes antérieurs.
