Abstract of the Disclosure A recombinant expression vector is constructed with a strong promoter element and a restriction endonuclease site positioned downstream from the promoter so that trans- cription initiation always occurs at a precise nucleotide. Following the conversion of viral RNA molecules to cDNA by the use of reverse transcriptase, the cDNA molecules can be transcribed in vitro into RNA. Viral RNA molecules transcribed in vitro from cDNA molecules are infectious, provided that transcription initiation and transcription termination are essentially at the correct nucleotides.
