The well-differentiated airway epithelium is the principal target tissue for gene therapy for the treatment of CF. However, recent studies have shown that gene delivery vehicles, such as cationic lipid:DNA complexes, can be inefficient at binding to and internalizing into polarized epithelial cells. The present invention provides a method to improve gene therapy by using a compound capable of disrupting tight junctions. In the practice of the invention, the transfection of a biologically active molecule by a cationic amphiphile:biologically active molecule complex or other lipid or viral or nonviral vectors is improved by treating the cells with a class of compounds known in the art as absorption enhancers or tight junction disrupting compounds.

L'épithélium des voies aériennes bien différencié est le principal tissu cible pour une thérapie génique destinée à traiter la mucoviscidose. Toutefois, des études récentes ont montré que les véhicules de distribution de gène, tels que les complexes lipides/ ADN cationiques, peuvent être inefficaces pour les lier à des cellules épithéliales polarisées et les transformer en de telles cellules. L'invention concerne un procédé permettant d'améliorer la thérapie génique, au moyen d'un composé capable d'interrompre des jonctions serrées. Dans la pratique de l'invention, la transfection d'une molécule biologiquement active par un complexe moléculaire biologiquement actif/amphiphile cationique, un autre lipide ou des vecteurs viraux ou non viraux, est améliorée par traitement des cellules avec des composés d'une classe connue dans la technique, sous forme de composés renforceurs d'absorpsion ou interrupteurs de jonction serrée.
