Transgene constructs for generating transgenic animals, wherein the transgene encodes a gene product which modulates transcription of a hypertrophy- sensitive gene, are provided. Further provided are recombinant vectors comprising the transgenes of the invention. Further provided are transgenic animals generated using the transgene constructs. Further provided are enzyme- based, cell-based, and whole-animal-based assays for detecting substances having therapeutic activity toward cardiac hypertrophy. Further provided are compositions comprising substances which modulate levels of active product of a hypertrophy-sensitive gene. Further provided are methods of treating cardiac hypertrophy.
Cette invention se rapporte à des transgènes de synthèse servant dans la production d'animaux transgéniques, un tel transgène codant un produit génique qui module la transcription d'un gène sensible à l'hypertrophie. Cette invention se rapporte en outre à des vecteurs de recombinaison comprenant ces transgènes, à des animaux transgéniques utilisant ces transgènes de synthèse, à des dosages ayant recours à des enzymes, à des cellules ou à l'animal entier pour détecter des substances produisant une action thérapeutique sur l'hypertrophie cardiaque, à des compositions contenant des substances qui modulent les niveaux de produit actif d'un gène sensible à l'hypertrophie, ainsi qu'à des procédés servant à traiter l'hypertrophie cardiaque.
