The invention provides cloned transgenic ungulates (e.g., bovines) in which prion protein activity is reduced by one or more genetically engineered mutations. Desirably, these transgenic bovines are also genetically modified to express xenogenous (e.g., human) antibodies. Because of their resistance to prion-related diseases such as bovine spongiform encephalopy (also known as mad cow disease), these bovines are a safer source of human antibodies for pharmaceutical uses and safer source of agricultural products.
L'invention concerne des ongulés transgéniques, clonés (par exemple des bovins), chez lesquels l'activité de la protéine prion est réduite par une ou plusieurs mutations génétiquement modifiées. De préférence, lesdits bovins transgéniques sont également génétiquement modifiés, de manière à exprimer des anticorps xénogènes (par exemple des anticorps humains). Du fait de leur résistance aux maladies associées au prion telles que l'encéphalopathie spongiforme bovine (connue également sous le nom de maladie de la vache folle), les bovins sont une source plus sûre d'anticorps humains pour des utilisations pharmaceutiques et une source plus sûre de produits agricoles.
