Provided are methods of designing a putative inhibitor of a 5'- methylthioadenosine/S-adenosylhomocysteine nucleosidase. The methods comprise designing a chemically stable compound that resembles the charge and geometry of the 5'-methylthioadenosine/S-adenosylhomocysteine nucleosidase transition state. Also provided are methods of inhibiting 5'-methylthioadenosine/S- adenosylhomocysteine nucleosidases using the inhibitors found by the above methods.
La présente invention porte sur des procédés permettant de concevoir un inhibiteur putatif d'une 5'-méthylthioadénosine/S-adénosylhomocystéine nucléosidase. Les procédés consistent à concevoir un composé chimiquement stable qui rappelle la charge et la géométrie de l'état de transition de la 5'-méthylthioadénosine/S-adénosylhomocystéine nucléosidase. Cette invention concerne également des procédés d'inhibition de 5'-méthylthioadénosine/S-adénosylhomocystéine nucléosidases utilisant les inhibiteurs trouvés au moyen des procédés susmentionnés.
