Provided is a method for selecting a chemotherapeutic agent for treating cancer, which method comprises: (a) testing a sample comprising p53 mutant cells, or an extract therefrom for the level of expression of Cyclin D1 or for the abundance of Cyclin D1 protein; and (b) if Cyclin D1 is over-expressed, and/or Cyclin D1 protein is present at elevated levels, selecting for treatment a chemotherapeutic agent comprising a taxane; (c) if Cyclin D1 is not overexpressed and/or Cyclin D1 protein is substantially not present at elevated levels, selecting for treatment a chemotherapeutic agent comprising an agent other than a taxane. Also provided is a kit for selecting a chemotherapeutic agent for treatment, which kit comprises: (a) a means for identifying p53 mutant cells; and (b) a means for testing for the level of expression of Cyclin D1 or for the abundance of Cyclin D1 protein in cells or in a sample therefrom.
L'invention concerne une technique permettant de sélectionner un agent de chimiothérapie. Cette technique consiste (a) à tester un échantillon comprenant des cellules mutantes p53, ou un extrait de celles-ci, pour déterminer le niveau d'expression de la cycline D1 ou l'abondance de la protéine de la cycline D1, (b) à sélectionner, en vue du traitement, un agent chimiothérapique comprenant un taxane, si la cycline D1 est surexprimée, et/ou si une protéine de la cycline D1 est présente à des niveaux élevés; (c) à sélectionner, en vue du traitement, un agent chimiothérapique différent du taxane, si la cycline n'est pas surexprimée et/ou si une protéine de la cycline D1 n'est pas sensiblement présente à des niveaux élevés. L'invention concerne également un kit de sélection d'un agent de chimiothérapie. Ce kit comprend (a) des moyens d'identification des cellules mutantes p53; et (b) des moyens de recherche du niveau d'expression de la cycline D1 ou de l'abondance d'une protéine de la cycline D1 dans des cellules ou dans un échantillon de celles-ci.
