Markedly enhanced activity of pacemaker (hyperpolarization-activated, cation- nonselective, HCN) ion channels governs spontaneous firing in sensory cells of allodynic rats. An HCN ion channel specific blocker, ZD7288, dose-dependently and completely suppresses allodynia. Nerve injury increases the population of large DRG neurons expressing a high density of Ih and modulates HCN mRNA expression. New methods of treating pain by targeting HCN pacemaker channels are developed. In addition, new methods for identifying compositions useful for treating pain are disclosed.
Une amélioration notable de l'activité des canaux ioniques (activés par hyperpolarisation, non sélectifs des cations, HCN) du pacemaker dirige la décharge spontanée dans les cellules sensorielles des rats allodyniques. Un bloqueur spécifique aux canaux ioniques HCN, ZD7288, supprime l'allodynie complètement et en fonction de la dose. Une lésion de nerfs fait augmenter la population de grands neurones DRG exprimant une densité élevée d'Ih et module l'expression d'ARNm de HCN. On propose de nouveaux procédés pour traiter la douleur en ciblant les canaux de pacemaker HCN. En outre, l'invention concerne de nouveaux procédés pour identifier des compositions utiles dans le traitement de la douleur.
