The present invention relates generally to a method of treatment and/or prevention of hypersensitivity including anaphylaxis in patients receiving replacement therapy molecules that elicit an IgE-mediated response by administering an anti-IgE antibody or a binding fragment thereof. Anti-IgE antibodies inhibit IgE-mediated allergic reactions in the mammals and can also lower the risk of an anaphylactic reaction to a replacement therapy molecule. The administration of anti-IgE antibodies over time also downregulates the high-affinity IgE receptor, further decreasing the risk of hypersensitivity and/or anaphylaxis. The anti-IgE antibody binds to circulating or serum IgE and/or membrane form IgE on B-cells, but not to IgE bound to mast cells or basophils, as this may cause crosslinking. This method also reduces the dose and/or frequency of administration of the replacement therapy molecule by reducing or eliminating neutralizing IgE antibodies specific for the replacement therapy molecule.
L'invention concerne une méthode de traitement et/ou de prévention de l'hypersensibilité comprenant l'anaphylaxie chez des patients recevant des molécules de thérapie de remplacement qui élicitent une réponse médiée par l'IgE, grâce à l'administration d'un anticorps anti-IgE ou d'un de leurs fragments de liaison. Des anticorps anti-IgE inhibent les réactions allergiques médiées par l'IgE chez les mammifères et peuvent également diminuer le risque d'une réaction anaphylactique sur une molécule de thérapie de remplacement. L'administration des anticorps anti-IgE dans le temps régule par le bas le récepteur de l'IgE à grande affinité, ce qui permet de diminuer le risque d'hypersensibilité et/ou d'anaphylaxie. L'anticorps anti-IgE fixe l'IgE circulant ou le sérum et/ou l'IgE sous forme de membrane sur des cellules B, mais pas des liaisons d'IgE sur des cellules mastocytes ou des basophiles, cela entraînant une réticulation. Ledit procédé permet également la réduction de la dose et/ou de la fréquence d'administration de la molécule de thérapie de remplacement par réduction ou élimination des anticorps neutralisant l'IgE spécifiques pour la molécule de thérapie de remplacement.
