A method for treating diabetes mellitus by administering composition providing a gastrin/CCK receptor ligand, e.g. a gastrin, and an EGP receptor ligand, e.g. TGF.alpha., in an amount sufficient to effect differentiation of pancreatic islet precursor cells to mature insulin-secreting cells. The composition can be administered systemically or expressed in situ by cells transgenically supplemented with one or both of a gastrin/CCK receptor ligand gene, e.g. a preprogastrin peptide precursor gene and an EGF receptor ligand gene, e.g. a TGF.alpha. gene.
L'invention concerne un procédé de traitement du diabète sucré, consistant à administrer une composition contenant un ligand au récepteur de gastrine/cystokinine (CCK), par exemple une gastrine, et un ligand au récepteur d'un facteur de croissance épidermique, par exemple le facteur de croissance transformant .alpha., en une quantité suffisante pour que se fasse la différenciation de cellules précurseurs d'îlots pancréatiques en cellules mûres sécrétant de l'insuline. Cette composition peut être administrée par voix générale ou exprimée in situ par des cellules transgéniquement complétées par au moins un des gènes suivants: un gène de ligand au récepteur de gastrine/CCK, par exemple un gène précurseur peptidique de préprogastrine, et un gène de ligand au récepteur d'un facteur de croissance épidermique, par exemple un gène du facteur de croissance transformant .alpha..
