Examples and embodiments of the present invention teach and disclose, inter alia, that the previously known extracellular matrix metallproteinase inducer protein (EMMPRIN), also known as CD147, is a signal-transducing cellular receptor for cyclophilin (CyP). Accordingly, there is disclosed methods for the treatment of HIV-infection, AIDS, AIDS-related disorders., rheumatoid arthritis (RA), connective tissue disorders, cancer or any condition characterized by local or systemic CyP release, synthesis or binding using the CyP/EMMPRIN interaction as a novel therapeutic intervention target. There is further disclosed pharmaceutical compositions including, anti-EMMPRIN antibodies, and soluble EMMPRIN proteins and peptides that act by inhibiting the CyP/EMMPRIN interaction. The present invention further provides screening assays for the identification of compounds that inhibit the CyP/EMMPRIN interaction.
Des exemples et des modes de réalisation de l'invention montrent, entre autres, que la protéine inductrice de métalloprotéinase matricielle extracellulaire (EMMPRIN), déjà connue, appelée également CD147, constitue un récepteur cellulaire transducteur de signal pour la cyclophiline (CyP). L'invention concerne des méthodes de traitement de l'infection par VIH, du sida, de troubles liés au sida, de la polyarthrite rhumatoïde (RA), des troubles tissulaires associés, du cancer ou de tout état caractérisé par une sécrétion, synthèse ou fixation locale ou systémique de CyP, lesdites méthodes utilisant l'interaction CyP/EMMPRIN comme nouvelle cible d'intervention thérapeutique. L'invention concerne en outre des compositions pharmaceutiques comprenant des anticorps anti-EMMPRIN, et des protéines et peptides EMMPRIN solubles qui agissent par inhibition de l'interaction CyP/EMMPRIN. L'invention concerne également des méthodes de criblage pour identifier des composés qui inhibent l'interaction CyP/EMMPRIN.
