The invention concerns tricyclic .NU.-heteroaryl-carboxamide derivatives containing a benzimidazole unit, of general formula (I) wherein: A represents, with the C-N bond of the benzimidazole unit to which it is bound, a 4- to 7-membered monocyclic heterocycle or a monocyclic heteroaryl containing from one to three heteroatoms selected among O, S or N, including the nitrogen atom of the benzimidazole unit; P represents a 8-, 9-, 10-, 11-membered bicyclic heterocycle or a bicyclic heteroaryl, comprising 1 to 6 heteroatoms selected among N, O or S; R1 represents on to four atoms or groups, identical or different; Y represents an oxygen or sulfur atom; R2 represents a hydrogen atom, a halogen atom, a C1C6-alkyl, C3-C7- cycloalkyl, C3-C7-cycloalkyl-C1-C3-alkylene, C1C6-fluoroalkyl or C1C6-alkoxy group; R3 represents from one to three atoms or groups, identical or different; in base or acid addition salt state as well as in hydrate or solvate form. The invention also concerns a method for preparing the compounds and their therapeutic use.
L'invention concerne les dérivés de N-hétéroaryl-carboxamides tricycliques contenant un motif benzimidazole, de formule générale (I) dans laquelle A représente, avec la liaison C-N du motif benzimidazole auquel il est fusionné, un hétérocycle monocyclique ou un hétéroaryle monocyclique de 4 à 7 chaînons, contenant de un à trois hétéroatomes choisis parmi O, S ou N , y compris l'atome d'azote du motif benzimidazole ; P représente un hétérocycle bicyclique ou hétéroaryle bicyclique à 8-, 9-, 10- ou 11 chaînons, comprenant de 1 à 6 hétéroatomes sélectionnés parmi N, O et S ; Ri représente de un à quatre atomes ou groupes, identiques ou différents ; Y représente un atome d'oxygène ou de soufre ; R2 représente un atome d'hydrogène, un atome d'halogène, un groupe C1C6-alkyle, C3-C7- cycloalkyle, C3-C7-cycloalkyle-C1-C3- alkylène, C1C6-fluoroalkyle ou C1C6-alcoxy ; R3 représente de un à trois atomes ou groupes, identiques ou différents ; à l'état de base ou de sel d'addition à un acide, ainsi qu'à l'état d'hydrate ou de solvate. L'invention concerne également un procédé de préparation des composés et leur application en thérapeutique.
