A method and associated kit for analysing genomic DNA in a sample, said method comprising: providing first and second oligonucleotide primers, said first oligonucleotide primer being a 5' variation generator, and comprising a repeat sequence and at least one nucleotide inconsistent with the repeat pattern, said first oligonucleotide primer having at least one nucleotide positioned on the first oligonucleotide's 5' end, and said second oligonucleotide primer being a 3' fragment generator starting within such a genetic distance that amplification of the genomic DNA can be performed; conducting a nucleic acid amplification on said genomic DNA in the sample at a relatively low annealing temperature using both the first and second oligonucleotide primers, said amplification being conducted under conditions such that neither the first nor the second oligonucleotide primer alone can amplify DNA, thus producing DNA fragments based on repeat sequences on one end of the genomic DNA and other sequences based on the opposite end of the genomic DNA; and analysing the amplified products thus produced to determine the length of a repeat sequence found in said genomic DNA.
L'invention concerne une méthode et un matériel associé d'analyse de l'ADN génomique dans un échantillon, ladite méthode consistant à produire des première et seconde amorces oligonucléotidiques, ladite première amorce oligonucléotidique étant un générateur de variation en 5', et comprenant une séquence répétée ainsi qu'au moins un nucléotide incohérent avec le type de répétition, ladite première amorce oligonucléotidique ayant au moins un nucléotide positionné sur l'extrémité 5' du premier oligonucléotide, et ladite seconde amorce oligonucléotidique étant un générateur de fragments 3' commençant à l'intérieur d'une distance génétique telle que l'amplification de l'ADN génomique puisse être exécutée, à exécuter une amplification d'acide nucléique sur ledit ADN génomique dans l'échantillon à une température de recuit relativement basse, à l'aide à la fois de la première et de la seconde amorce oligonucléotidique, ladite amplification étant exécutée dans des conditions telles que ni la première ni la seconde amorce oligonucléotidique seule ne puisse amplifier l'ADN, produisant ainsi des fragments d'ADN basés sur des séquences répétées à une extrémité de l'ADN génomique et d'autres séquences basées sur l'extrémité opposée de l'ADN génomique, et à analyser les produits amplifiés ainsi produits afin de déterminer la longueur d'une séquence répétée trouvée dans ledit ADN génomique.
