1,4-Dihydropyridines have been found to counter several processes which play a role in the development of atherosclerotic vascular lesions, such as myocytes proliferation and migration, cholesterol metabolism in macrophages and oxidative modification of low density lipoproteins. They are therefore useful in the manufacture of medicaments for preventing, arresting and reversing atherosclerotic degradation in the arterial walls of humans. The preferred 1,4- dihydropyridines for this purpose are lercanidipine, (S)-lercanidipine and (R)- lercanidipine.
On a découvert que les 1,4 dihydropyridines contrent plusieurs processus impliqués dans le développement des lésions vasculaires athéroscléreuses, telles que la prolifération et la migration de myocytes, le métabolisme du cholestérol dans les macrophages et la modification oxydative des lipoprotéines de basse densité. Ces dérivés s'avèrent par conséquent d'utilité pour fabriquer des médicaments permettant de prévenir, de circonscrire et d'inverser la dégradation athérosclérotique dans les parois artérielles chez l'homme. Les 1,4 dihydropyridines préférées utilisées à cette fin sont la lercanidipine, la (S)-lercanidipine et la (R)-lercanidipine.
