The invention relates to methods and kits for detecting enzyme activity using bioluminescence. In particular, it relates to a novel assay system with increased light yield for a sensitive and convenient detection of luciferase activity, such as luciferase reporter enzyme activity. Provided is a method of detecting luciferase activity in a sample, comprising incubating the sample in the presence of luciferin and ATP to allow the generation of a light signal, wherein said light signal is enhanced by performing the incubation in a reaction mixture comprising phosphate and ammonium ions, and measuring the light signal. The invention also relates to kits for use in such method.
La présente invention se rapporte à des procédés et à des kits permettant de détecter une activité enzymatique à l'aide de la bioluminescence. L'invention a trait en particulier à un nouveau système de dosage à rendement lumineux amélioré permettant de détecter avec précision et facilité l'activité luciférase, telle que l'activité de l'enzyme rapporteur luciférase. L'invention concerne un procédé permettant de détecter l'activité luciférase dans un échantillon, qui consiste à procéder à l'incubation de l'échantillon en présence de luciférine et d'ATP afin de permettre la génération d'un signal lumineux, ledit signal lumineux étant renforcé lorsque l'incubation est réalisée dans un mélange réactionnel contenant des ions phosphate et ammonium, et à mesurer le signal lumineux. L'invention a également trait à des kits destinés à être utilisés dans le cadre d'un tel procédé.
