The present invention provides methods for direct killing of cancer cells using anti-.alpha.5.beta.1 antibodies. Generally, the method comprises contacting a cancer cell that expresses .alpha.5.beta.1 on its surface with an anti-.alpha.5.beta.1 antibody, and thereby inducing the death of the cancer cell. The methods of the invention may be employed at an early stage of cancer development in a patient to prevent tumor establishment. In addition, the methods may be used to treat previously formed tumors especially in cancer that have not proven susceptible to anti-angiogenesis therapy. The methods may be employed as a combination therapy of anti-.alpha.5.beta.1 antibodies together with cancer chemotherapeutic agents or other molecular-based cancer therapeutic agents.
L'invention concerne des méthodes visant à tuer directement des cellules cancéreuses au moyen d'anticorps anti-.alpha.5.beta.1. La méthode selon l'invention consiste généralement à mettre en contact une cellule cancéreuse exprimant .alpha.5.beta.1 sur sa surface avec un anticorps anti-.alpha.5.beta.1, ce qui a pour effet d'induire la mort de la cellule cancéreuse. Les méthodes selon l'invention peuvent être mises en oeuvre à un stade précoce du développement d'un cancer chez un patient, de sorte à prévenir l'établissement de la tumeur. Ces méthodes peuvent également être mises en oeuvre pour traiter des tumeurs déjà formées, notamment dans le cas d'un cancer qui ne s'est pas révélé sensible à un traitement anti-angiogenèse. Ces méthodes peuvent être mises en oeuvre sous la forme d'une polythérapie associant des anticorps anti-.alpha.5.beta.1 et des agents chimiothérapeutiques anticancéreux ou d'autres agents anticancéreux à base moléculaire.
