The invention is directed to methods for in vitro DNA synthesis catalysed by a DNA polymerase using cyclodex-trins. The invention also relates to methods, compositions and kits comprising cyclodextrins for the amplification of a nucleic acid. The use of cyclodextrins improves the specificity, sensibility and/or yield of the amplification reaction. The invention is re-lated more particularly to kits, compositions and methods for carrying out PCR reactions comprising a cyclodextrin.
