The invention relates to a method for screening compounds, which inhibit MetAP2 activity, by providing a cellular system or a sample thereof being capable of expressing MetAP2 and/or EEF1A, incubating at least a portion of the system with compounds to be screened, and detecting MetAP2 inhibition by determining EEF1A with N-terminal methionine residue (MetEEFIA). Another object of the invention concerns a method for monitoring physiological and/or pathological conditions, which are caused, mediated and/or propagated by MetAP2 activity, by administering an effective amount of at least a single compound to a mammal in need of such treatment and determining MetEEFIA in a biological sample withdrawn from the mammal. The invention also relates to the use of EEF1A as biomarker.
La présente invention concerne un procédé permettant le criblage de composés inhibiteurs de lactivité de MetAP2, par la fourniture dun système cellulaire ou dun échantillon de celui-ci capable dexprimer MetAP2 et/ou eEF1A,, lincubation dau moins une partie du système avec les composés à cribler, et la détection dinhibition de MetAP2 par la détermination dEEF1A avec un résidu de méthionine à lextrémité N (MetEEFIA). Linvention concerne également un procédé permettant le contrôle de conditions physiologiques et/ou pathologiques, qui sont provoquées, médiées et/ou propagées par lactivité de MetAP2, par ladministration dune quantité efficace dau moins un composé unique à un mammifère qui a besoin dun tel traitement et la détermination de MetEEFIA dans un échantillon biologique prélevé du mammifère. Linvention concerne également lutilisation dEEF1A comme biomarqueur.
