The use of fibroblast growth factor (FGF)-18 protein, certain of its downstream target genes and respective expressed proteins, in particular sonic hedgehog (Shh), Shh protein, .beta.-catenin, .beta.-catenin protein, and the Wnt family of proteins that stimulate .beta.-catenin, and the respective nucleotide sequences encoding this protein, particularly for inducing cartilage formation, particularly for the purpose of generating, repairing, reconstructing, or de novo formation of, cartilaginous tissue. Therapies for which FGF-18 and the target proteins are useful include repair and reconstruction of various tissues in conducting airways such as the trachea, bronchi, lung and larynx caused by, for example, tracheal-bronchial abnormalities, tracheal-laryngo or bronchial malaria. Other therapies for which FGF-18 and the target proteins would be useful include other cartilaginous tissues, such as those of joint and skeletal tissue caused by, for example, arthritis and meniscus abnormalities in joints.
Cette invention se rapporte à l'utilisation de la protéine du facteur de croissance des fibroblastes 18 (FGF-18), de certains de ses gènes cibles en aval et des protéines exprimées respectives, en particulier le morphogène Sonic Hedgehog (Shh), la protéine Shh, la .beta.-caténine, la protéine de .beta.-caténine et la famille Wnt de protéines qui stimulent la .beta.-caténine, et des séquences nucléotidiques respectives codant cette protéine, notamment pour induire la formation de cartilages, en particulier en vue de générer, réparer, reconstruire ou former de nouveau des tissus cartilagineux. Les thérapies pour lesquelles ce facteur FGF-18 et ces protéines cibles sont utiles sont notamment la réparation et la reconstruction de divers tissus dans les voies respiratoires, telles que la trachée, les bronches, les poumons et le larynx, après des lésions causées par exemple par des anomalies trachéo-bronchiques, par une trachéo-laryngomalacie ou par une bronchomalacie. D'autres thérapies pour lesquelles ce facteur FGF-18 et ces protéines cibles seraient utiles concernent d'autres tissus cartilagineux tels que ceux des articulations et d'autres os du squelette, à la suite de lésions causées notamment par des arthrites et des anomalies de ménisques dans des articulations.
