A strategy for drug-screening is based on cells that are isogenic except for a gene of interest. Each cell can be tranfected with a vector that encodes a different fluorescent protein that can be differentially detected to monitor cell growth. Co-culture of both cells allows facile screening for compounds with selective toxicity towards a gene of interest. The drug screening is broadly applicable for mining therapeutic agents targeted to specific genetic alterations responsible for cancer development.

La présente invention se rapporte à une méthode de criblage de médicaments fondée sur des cellules qui sont isogéniques sauf pour un gène d'intérêt. Chaque cellule peut être transfectée par un vecteur codant une protéine fluorescente différente qui peut être détectée de manière différentielle aux fins de la surveillance de la croissance cellulaire. La co-culture des deux cellules facilite le criblage de composés présentant une toxicité sélective vis-à-vis d'un gène d'intérêt. Ce criblage de médicaments est largement applicable à la sélection d'agents thérapeutiques ciblant des altérations génétiques spécifiques responsables du développement d'un cancer.
