The invention provides a method of nonenzymatic ligation of a nucleic acid. The method consists of contacting a polynucleotide-3' phosphorothiolate with aan acceptor polynucleotide under conditions that allow formation of a phosphodiester bond between the polynucleotide. The invention also provides methods of molecular cloning. In one embodiment, the method consists of contacting an insert conprising a polynucleotide-3' phosphorothiolate with an acceptor vector under conditions that allow formation of a phosphodiester bond between the insert and the acceptor vector to generate a vector comprising an insert polynucleotide. The invention further provides a compound consisting of a polynucleotide-3' phosphorothiolate and a kit containing a polynucleotide-3' phosphorotiolate. Also provided is a method of ligating a nucleic acid using a non-sequence specific topoisomerase.

L'invention concerne une méthode de ligation non enzymatique d'un acide nucléique. Ladite méthode consiste à mettre un polynucléotide-3' de phosphorothiolate en contact avec un polynucléotide accepteur dans des conditions permettant la formation d'une liaison phosphodiester entre le polynucléotide-3' de phosphorothiolate et le polynucléotide accepteur. L'invention concerne également des méthodes de clonage moléculaire. Dans un mode de réalisation, la méthode consiste à mettre un insert comprenant un polynucléotide-3' de phosphorothiolate en contact avec un vecteur accepteur dans des conditions permettant la formation d'une liaison phosphodiester entre l'insert et le vecteur accepteur afin de produire polynucléotide d'insertion. L'invention concerne, en outre, un composé et un kit contenant un polynucléotide-3' de phosphorotiolate. Elle concerne enfin une méthode de ligation d'acide nucléique utilisant une topoisomérase spécifique non séquencée.
