Methods of enabling or improving the ability of a hydrogel to swell in the stomach of an animal and/or increasing the amount of time said hydrogel remains swollen in the stomach are described herein. In one embodiment, a polymer is administered in combination with one or more pH modifying agents which raise and maintain the pH of the micro environment of the polymer and/or the stomach in order inducing swelling in the polymer. The polymer can be a homopolymer, a copolymer, or a polymer blend or composite. In one embodiment, the polymer is a superabsorbent polymer ("SAP"). The polymers can also be administered with one or more active agents, such as appetite suppressants. The pH modifying agent and/or the active agent can be administered simultaneously with the polymer in the same dosage form, simultaneously with the polymer in separate dosage forms, or sequentially. The compositions are formulated for oral administration. The formulation can include drugs for delivery to the stomach, such as antibiotics, or the hydrogel can be used as a filler, for example, for obesity control. The formulation an also be used to enhance gastric retention, for example, for controlled drug delivery. Methods of delivering a drug are also described herein, along with medicaments for carrying out the methods of the present invention.
L'invention concerne des procédés permettant ou améliorant l'aptitude d'un hydrogel à gonfler dans l'estomac d'un animal et/ou augmentant la durée pendant laquelle ledit hydrogel reste gonflé dans l'estomac. Dans un mode de réalisation, un polymère est administré en association avec un ou plusieurs agents de modification du pH qui augmentent et maintiennent le pH du microenvironnement du polymère et/ou de l'estomac afin d'induire un gonflement du polymère. Le polymère peut être un homopolymère, un copolymère ou un mélange ou composite de polymères. Dans un mode de réalisation, le polymère est un polymère super absorbant ('SAP'). Les polymères peuvent également être administrés avec un ou plusieurs agents actifs, tels que des anorexigènes. L'agent de modification du pH et/ou l'agent actif peut être administré simultanément avec le polymère dans la même forme de dosage, simultanément avec le polymère dans des formes de dosage séparées, ou séquentiellement. Les compositions sont formulées pour une administration par voie orale. La formulation peut comporter des médicaments pour une administration à l'estomac, tels que des antibiotiques, ou l'hydrogel peut être utilisé comme charge, par exemple pour lutter contre l'obésité. La formulation peut également être utilisée pour améliorer la rétention gastrique, par exemple pour une administration de médicaments contrôlée. L'invention concerne également des procédés d'administration de médicaments, conjointement avec les médicaments destinés à mettre en AEuvre les procédés de la présente invention.
