The Streptomycetes plasmid pSG5 is temperature-sensitive. It is thus possible with the aid of the pSG5 replicon to prepare temperature-sensitive hybrid plasmids. The latter are suitable for screening for DNA sections which are homologous with DNA segments of the hybrid plasmid.
