An improved method of amplifying target nucleic acids and particularly, to a method of alleviating the inhibitory effect of certain samples on ligase chain reaction amplifications. The relief of inhibition is accomplished by adding to the amplification mixture a polyamine reagent such as spermidine, in effective amounts, typically 1 to 4 nM. In other embodiments, the presence of spermidine allows for amplification to proceed in the presence of reduced concentrations of MgCl2 and ligase.

Procédé amélioré d'amplification d'acides nucléiques cibles et en particulier procédé d'atténuation des effets inhibiteurs de certains échantillons lors de réactions d'amplification en chaine par ligases. L'atténuation de l'inhibition s'obtient par adjonction au mélange d'amplification d'une quantité efficace d'un réactif de type polyamine tel que la spermidine, par exemple de 1 à 4 mM. Dans des variantes, la présence de la spermidine permet l'amplification en présence de concentrations réduites de MgCl¿2? et de ligase.
