The present invention is directed to a method for preparing an expression vector encoding a tailored recombinase, wherein said tailored recombinase recombines asymmetric target sites within the LTR of proviral DNA of a retrovirus inserted into the genome of a host cell and is useful as means for excising the provirus from the genome of the host cell. The present invention further relates to an in vitro-method of optimising the treatment of a retroviral infection of a subject and to the use of tailored recombinases for the preparation of pharmaceutical compositions for reducing the viral load in a subjected infected by a retrovirus.
La présente invention porte sur un procédé de préparation d'un vecteur d'expression codant pour une recombinase personnalisée. Ladite recombinase personnalisée recombine des sites cibles asymétriques dans la longue répétition terminale (LTR) d'un ADN proviral d'un rétrovirus inséré dans le génome d'une cellule hôte et est utile comme moyen pour exciser le provirus du génome de la cellule hôte. La présente invention porte en outre sur un procédé in vitro d'optimisation du traitement d'une infection rétrovirale d'un sujet et sur l'utilisation de recombinases personnalisées pour la préparation de compositions pharmaceutiques afin de réduire la charge virale chez un sujet infecté par un rétrovirus.
