ABSTRACT OF THE DISCLOSURE: The invention is concerned with a method for the determination of the concentration of tri-iodo thyronine in a sample. The method of the invention comprises extracting the tri-iodo thyronine of the sample being assayed by means of an insolubilized anti-T3 antibody derived from serum and tri-iodo thyronine; contacting the extracted tri-iodo thyronine in a column with a sensibilized enzyme consisting of an enzyme that is covalently bonded with tri-iodo thyronine and is derived from carbonic anhydrase and tri-iodo thyronine; hydrolyzing the effluent from the column with p-nitrophenyl acetate; and spectrophotometrically determining the concentration of tri-iodo thyronine by measuring the absorption of p-nitro- phenyl acetate hydrolyzed washings from the column in comparison with a standard. The invention enables one to dose tri-iodo thyronine in extremely small amount, in the order of the nano- gram and under.
