Expression vectors are disclosed comprising intronless genes containing one or more near consensus splice sequences and one or more copies of a viral cis- acting post-transcriptional regulatory element which is transcribed along with the gene and causes export of the gene transcript from the nucleus into the cytoplasm of the cell. In a preferred embodiment, the vectors are targeted for delivery to specific cells in the form of a molecular complex made up of the plasmid releasably linked to a nucleic acid binding agent and a ligand which binds to a component on the surface of a cell. Use of viral cis-acting post- transcriptional regulatory elements as disclosed can increase expression of intronless genes with near-consensus splice sites.

L'invention concerne des vecteurs d'expression comprenant des gènes sans introns, contenant une ou plusieurs séquences d'épissage proches du consensus et une ou plusieurs copies d'un élément régulateur viral post-transcription agissant en cis. Celui-ci est transcrit avec le gène et entraîne l'exportation du produit de transcription du gène depuis le noyau dans le cytoplasme de la cellule. Dans un mode de réalisation préféré, les vecteurs sont ciblés pour être fournis à des cellules spécifiques sous forme d'un complexe moléculaire composé du plasmide lié, de manière libérable, à un agent de liaison de l'acide nucléique et d'un ligand qui se lie à un constituant sur la surface d'une cellule. L'invention concerne l'utilisation d'éléments régulateurs viraux post-transcription agissant en cis, qui permettent d'augmenter l'expression de gènes sans introns avec des sites d'épissage proches du consensus.
