The present invention relates to novel vaccine therapies, and prophylactic treatments of atherosclerotic diseases. Accordingly there is provided, immunogens comprising specific fragments or derivatives of Apolipoprotein C- III (ApoCIII). The vaccines of the present invention, comprising said immunogens, are potent in the prevention, or reduction, of atherosclerotic plaque formation over prolonged periods of time, thereby reducing the potential of atheroslerosis leading to coronary or cerebrovascular disease. Also provided are methods of treating or preventing atherosclerosis by active vaccination, or passive vaccination through administration to a patient of an antibody that is capable of binding to the specific fragments of ApoCIII. Specific monoclonal antibodies and their use in therapy of atherosclerosis is provided. There is further provided the use of the immunogens of the present invention in medicine, and methods of their production. The fragments of ApoCIII which form the basis of the immunogens of the present invention, and also the targets for passive immunotherapy, are encompased within the regions between amino acid numbers 45-76 and, particularly, 12-35 of the mature form of human ApoCIII.
La présente invention concerne de nouvelles thérapies à base de vaccins et des traitements prophylactiques de maladies athéroscléreuses. L'invention concerne, en conséquence, des immunogènes comprenant des fragments ou des dérivés spécifiques d'apolipoprotéine C-III (ApoCIII). Les vaccins susmentionnés comprenant lesdits immunogènes sont efficaces dans la prévention ou la diminution de la formation de la plaque d'athérosclérose sur des durées prolongées, réduisant ainsi la probabilité d'accidents coronaires ou cérébrovasculaires dûs à l'athérosclérose. L'invention concerne également des méthodes de traitement ou de prévention de l'athérosclérose par vaccination active ou vaccination passive consistant à administrer au patient un anticorps pouvant se fixer aux fragments spécifiques de l'ApoCIII. L'invention concerne des anticorps monoclonaux spécifiques et leur utilisation dans le traitement de l'athérosclérose. L'invention concerne également l'utilisation des immunogènes selon la présente invention dans un médicament, et leurs méthodes de production. Les fragments d'ApoCIII qui constituent la base des immunogènes de la présente invention, et également les cibles de l'immunothérapie passive, sont compris dans les régions situées entre les numéros d'acides aminés 45-76 et, notamment, 12-35 de la forme mûre de l'ApoCIII humaine.
