The present invention is directed to a method of selecting a clone that binds to human platelet glycoprotein Ib alpha using a human variable heavy chain and variable light chain immunoglobulin library. The invention is further directed to isolated nucleic acid molecules encoding a variable heavy chain or variable light chain region of an antibody, wherein the antibody binds to human platelet glycoprotein Ib alpha and inhibits aggregation of platelets. Expression vectors and host cells comprising the nucleic acid molecules are also provided, as well as methods for producing the variable heavy chain or the variable light chain region. An isolated variable heavy chain or variable light chain region of an antibody, wherein the antibody binds to human platelet glycoprotein Ib alpha and inhibits aggregation of platelets, is also provided. An antibody comprising the variable heavy chain or variable light chain regions is provided, as is a composition comprising the antibody and a carrier. The subject invention further provides a method of inhibiting aggregation of platelets, as well as a method of binding human platelet glycoprotein Ib alpha. A method of selecting a variable heavy chain or variable light chain region of an antibody is also provided.

L'invention concerne un procédé permettant de sélectionner au moyen d'une banque de régions variables de chaîne lourde et de chaîne légère d'immunoglobuline humaine, un clone qui se lie à la glycoprotéine plaquettaire humaine Ib alpha. L'invention concerne également des molécules d'acide nucléique isolées codant pour des régions variables de chaîne lourde ou de chaîne légère d'un anticorps. Cet anticorps se lie à la glycoprotéine plaquettaire humaine lb alpha et inhibe l'agrégation des plaquettes. L'invention concerne en outre des vecteurs d'expression et des cellules hôtes comprenant ces molécules d'acide nucléique ainsi que des procédés permettant de produire ces régions variables de chaîne lourde et de chaîne légère d'un anticorps, cet anticorps se liant à la glycoprotéine plaquettaire humaine Ig alpha et inhibant l'agrégation des plaquettes, ainsi qu'un anticorps comprenant ces régions variables de chaîne lourde et de chaîne légère et une composition comprenant l'anticorps et un porteur. L'invention concerne encore un procédé d'inhibition de l'agrégation plaquettaire ainsi qu'un procédé de fixation de la glycoprotéine Ib alpha humaine. Enfin l'invention concerne un procédé de sélection d'une région variable de chaîne lourde ou d'une région variable de chaîne légère d'un anticorps.
