A recombinant adenoviral vector encoding the human tissue factor pathway inhibitor (TFPI) gene is disclosed which is useful for transduction of vascular smooth muscle cells at a selected blood vessel site to provide local vascular expression of TFPI. A method of using the transduced hTFPI CDNA as an in vivo antithrombotic agent to provide localized production of hTFPI for protecting an at-risk site against thrombus deposition is also disclosed. Gene therapy using the new TFPI expression vector is also expected to deter the development of chronic vascular stenosis in blood vessels (arteries, veins, arteriovenous shunts, and endovascular grafts) and deterring intimal hyperplasia.

L'invention concerne un vecteur adénoviral recombinant codant pour le gène inhibiteur de la voie du facteur tissulaire (TFPI) humain, utile pour induire la transduction des cellules musculaires lisses vasculaires au niveau d'un site vasculaire sélectionné afin d'assurer l'expression vasculaire locale du TFPI. L'invention concerne également une méthode permettant d'utiliser l'ADNc de hTFPI transduit en tant qu'agent anti-thrombose in vivo afin d'induire la production localisée de hTFPI et de protéger ainsi un site à risque contre la formation de thrombus. Une thérapie génique utilisant ce nouveau facteur d'expression TFPI permettra de prévenir le développement d'une sténose vasculaire chronique dans les vaisseaux sanguins (artères, veines, shunts artério-veineux et greffons intravasculaires) et d'empêcher l'hyperplasie de l'intima.
