Described are vectors containing (a) a gene encoding (i) a CDK4/CDK6 inhibitor, preferably p16", or (ii) an RNA interfering with CDK4 and/or CDK6 expression and/or activity, under the control of an inducible promoter and (b) a gene encoding a transactivator protein for said promoter useful for treating neurodegenerative disorders. These vectors can be transferred into cells where they will exert a protective function to (i) prevent cell death or to (ii) slow down progression of cell death. These vectors can be used in therapeutic applications to prevent neurodegenerative disorders or to slow down their progression with therapeutic efficacy.
La présente invention concerne des vecteurs contenant (a) un gène codant pour (i) un inhibiteur CDK4/CDK6, de préférence p16INK4a, ou (ii) un ARN interférant avec l'expression et/ou l'activité de CDK4 et/ou CDK6, sous le contrôle d'un promoteur inductible, et (b) un gène codant pour une protéine de transactivateur pour ledit promoteur, utile dans le traitement de troubles neurodégénératifs. Lesdits vecteurs peuvent être transférés dans des cellules où ils exerceront une fonction protectrice afin de (i) prévenir la mort cellulaire ou de (ii) ralentir la progression de la mort cellulaire. Ces vecteurs peuvent être utilisés dans des applications thérapeutiques afin d'empêcher des troubles neurodégénératifs ou de ralentir leur progression avec une efficacité thérapeutique.
