A process for producing high purity 6,12-dideoyxerythromycin A using recombinant DNA technology is disclosed. The erythromycin producing strain, Saccharopolyspora erythraea, lacking the erythromycin C-12 and C-6 hydroxylases produces a mixture of 6,12-dideoxyerythromycin A and the precursor molecule, 6-deoxyerythromycin D. To achieve conversion of the precursor to the final product, a second copy of eryG is inserted into a non- essential region of the Sac. erythraea chromosome resulting in high purity 6,12-dideoxyerythromycin A.
Procédé de production de didésoxyérythromycine A de grande pureté reposant sur la technologie de recombinaison d'ADN. La souche de production d'érythromycine, Saccharopolyspora erythraea, qui ne contient pas les hydroxylases C-12 et C-6 de l'érythromycine, donne un mélange de didésoxyérythromycine A et de la molécule précurseur, 6-désoxyérythromycine D. Pour obtenir la conversion du précurseur en un produit final, une deuxième copie de eryG est insérée dans une région non essentielle du chromosome de Sac. Erythraea, permettant ainsi d'obtenir une 6,12-didésoxyérythromycine A de grande pureté.
