Lower eukaryote host cells in which the function of at least one endogenous gene encoding a chaperone protein, such as a Protein Disulphide Isomerase (PDI), has been reduced or eliminated and at least one mammalian homolog of the chaperone protein is expressed are described. In particular aspects, the host cells further include a deletion or disruption of one or more O-protein mannosyltransferase genes, and/or overexpression of an endogenous or exogenous Ca2+ ATPase. These host cells are useful for producing recombinant glycoproteins in large amounts and for producing recombinant glycoproteins that have reduced O-glycosylation.
La présente invention concerne des cellules hôtes eucaryotes inférieures dans lesquelles la fonction dau moins un gène endogène codant pour une protéine chaperon, telle quune protéine disulfure isomérase (PDI), a été réduite ou supprimée et dans laquelle au moins un homologue mammifère de la protéine chaperon est exprimé. Dans des aspects particuliers, les cellules hôtes comprennent en outre une suppression ou une dissociation dune ou plusieurs protéines O-mannosyl transférase, et/ou une surexpression dune Ca2+ ATPase endogène ou exogène. Ces cellules hôtes sont utiles dans la production de glycoprotéines recombinées en grandes quantités et dans la production de glycoprotéines qui présentent une O-glycosylation réduite.
