The present invention relates to viral inhibition, particularly HIV inhibition, by DNA sequences including multiple target response elements. The DNA construct of the present invention comprises a vector and a promoter operably linked to at least one target response element and, optionally, to other viral initibitory elements, so that the elements are inscribed in tandem. The DNA construct can be used in the treatment of viral infections, HIV in particular, by obtaining cells from an HIV-infected patient, transforming the cells with the construct and administering the transformed cells to the patient. The protective gene product will only be expressed if the cell becomes infected and a viral protein is made.
L'invention concerne l'inhibition virale, notamment l'inhibition du VIH, à l'aide de séquences d'ADN incluant des éléments de réaction cibles multiples. Le produit d'assemblage d'ADN de l'invention comprend un vecteur ainsi qu'un promoteur liés fonctionnellement à au moins un élément de réaction cible et, facultativement, à d'autres éléments inhibiteurs viraux, de manière que les éléments sont transcrits en tandem. Le produit d'assemblage d'ADN peut être utilisé dans le traitement d'infections virales, notamment du VIH, par l'obtention de cellules prélevées sur un patient infecté par le VIH, transformation des cellules à l'aide du produit d'assemblage et administration des cellules transformées audit patient. Le produit génique protecteur n'est exprimé que si la cellule devient infectée et une protéine virale est produite.
