The present disclosure provides nucleic acid constructs encoding one or more polypeptides containing at least one glycosaminoglycan chain, such as, but not limited to, a proteoglycan polypeptide, and methods for delivering to the site of a wound or cutaneous injury at least one nucleic acid construct encoding one or more such polypeptides, such that the expressed polypeptide is glycated by glycosaminoglycan chains through the normal physiological processes of the subject at the site of administration to produce a functional proteoglycan polypeptide for the healing of the wound or other cutaneous injury. The delivered nucleic acid construct is transcribed, translated and post-translationally modified by the addition of glycosaminoglycan chains (referred herein as "decoration" or "glycation") to produce a decorated polypeptide. The decorated polypeptide is then secreted from the cell in which it was produced to provide treatment of wounds and/or cutaneous injury in the subject and/or prevention of cutaneous injury in a subject.

La présente invention concerne des produits recombinants d'acide nucléique codant un ou plusieurs polypeptides contenant au moins une chaîne de glycosaminoglycane, telle qu'un polypeptide de protéoglycane notamment. L'invention se rapporte également à des procédés permettant de diffuser sur le site d'une plaie ou d'une blessure cutanée au moins un produit recombinant d'acide nucléique codant un ou plusieurs desdits polypeptides, de telle façon que le polypeptide exprimé soit glyqué par des chaînes de glycosaminoglycane par les processus physiologiques normaux du sujet au site de diffusion afin de produire un polypeptide de protéoglycane fonctionnel permettant la cicatrisation de la plaie ou autre blessure cutanée. Le produit recombinant d'acide nucléique diffusé est transcrit, traduit et modifié suite à cela par l'ajout de chaînes de glycosaminoglycane (opération dénommé = décoration = ou = glycation =) afin de produire un polypeptide décoré. Le polypeptide décoré est alors sécrété par la cellule dans laquelle il a été produit pour traiter les plaies et/ou les blessures cutanées chez le sujet et/ou prévenir les blessures cutanées chez un sujet.
