A liquid-droplet aerosol composition for delivery to a patient's respiratory tract is disclosed. The aerosol is formed by placing an aqueous .gamma.-IFN solution having stabilizing and dispers ing components against a plate having defined-size openings, and forcing the solution through the openings under conditions effective to form aqueous droplets having a volume mean diameter in one or more of a number of selected sizes in the 1-10 micron size range. The aerosol has the desired-size droplets and a .gamma.-I FN biological activity and molecular size distribution substantia lly the same as that of the aqueous .gamma.-IFN solution. Also disclosed is a method of administering to a selected region of a patient's respiratory tract, an amount of human .gamma.- IFN having a known, selected gamma-interferon biological activity and molecular size distribution.
L'invention concerne une composition aérosol à gouttelettes liquides destinée à l'administration dans les voies respiratoires d'un patient. On forme l'aérosol en plaçant une solution aqueuse de .gamma.-IFN renfermant des composants stabilisants et dispersants contre une plaque présentant des ouvertures dont la taille est définie, et en contraignant la solution à travers les ouvertures dans des conditions efficaces afin de former des gouttelettes aqueuses ayant un diamètre volumique moyen correspondant à un ou plusieurs nombres de dimensions sélectionnées dans la plage de dimensions comprise entre 1-10 micron. L'aérosol présente les gouttelettes de taille recherchée, une activité biologique de .gamma.-IFN et une administration moléculaire de taille sensiblement identiques à celles de la solution aqueuse de .gamma.-IFN. L'invention concerne également un procédé d'administration dans une région sélectionnée des voies respiratoires d'un patient d'une quantité de .gamma.-IFN humain présentant une activité biologique d'interféron gamma et une distribution de taille moléculaire sélectionnées et connues.
