Albumin, for example human albu- min, is expressed and secreted in yeast which has been mutated to lack the yeast aspartyl protease 3 (Yap3p) or its equiva- lent, thereby reducing the production of a 45kD albumin fragment. A further reduc- tion is achieved by additionally deleting the Kex2p function. Alternatively, a modified albumin is prepared which is not suscep- tible to Yap3p cleavage, for example hu- man albumin which is R410A, K413Q and K414Q.
L'albumine, d'origine humaine par exemple, est exprimée et sécrétée dans une levure qu'on a fait muter pour en supprimer d'aspartyle protéase 3 de levure (Yap3p) ou son équivalent, ce qui limite la production d'un fragment d'albumine de 45kD. Une limitation supplémentaire s'obtient en supprimant en plus la fonction Kex2p. On peut aussi préparer une albumine modifiée qui ne présente aucune tendance à la coupure de Yap3p, par exemple une albumine humaine telle que R410A, K413Q et K414Q.
