This invention relates to cyclic peptide derivatives of hirudin of the formula: (see fig. I) which are useful as anticoagulants. The cyclic peptides have a core sequence of approximately 12 amino acids in which there is an intra-cyclized chain that occurs partially through the alkyl chain of a D-amino acid. The modified peptides of this invention also possess significant anticoagulant activity which thereby allows a therapeutically significant adjunct to anticoagulant therapy.
