An in vivo method for the construction of randomized gene libraries and/or domain replacement in gene libraries by homologous recombination using a Kluyveromyces lactis killer toxin, in particular the (.gamma.-subunit of the K. lactis killer toxin, as negative selection marker is described. The use of the (.gamma.-subunit of K. lactis as negative selectable marker increases the percentage of randomized clones.
L'invention concerne un procédé in vivo pour la réalisation de génothèques randomisées et/ou le remplacement de régions dans des génothèques par recombinaison homologue à l'aide d'une toxine tueuse appelée Kluyveromyces lactis, notamment la sous-unité .gamma. de cette dernière, comme marqueur sélectif négatif. L'emploi de ladite sous-unité comme marqueur sélectif négatif permet d'augmenter le pourcentage de clones randomisés.
