A method for detecting a living microorganism, a damaged microorganism, a VNC microorganism and a dead microorganism in a sample by flow cytometry, the method comprising the steps of: a) treating a sample to be detected with an enzyme capable of degrading any non-microorganism cell, protein colloidal particle or fat present in the sample; b) treating the sample with a topoisomerase inhibitor and/or a DNA gyrase inhibitor; c) treating the sample given after the treatments in steps a) and b) with a nuclear staining agent; and d) detecting a microorganism or microorganisms treated with the nuclear staining agent in the sample by flow cytometry.
La présente invention concerne un procédé de détection de micro-organisme vivant, de micro-organisme endommagé, de micro-organisme VNC et de micro-organisme mort dans un échantillon par cytométrie de flux. Ledit procédé comprend les étapes suivantes : a) traiter un échantillon devant être décelé par une enzyme pouvant dégrader toute cellule qui n'est pas un micro-organisme, une particule colloïdale de protéine ou de la matière grasse présente dans l~échantillon ; b) traiter l'échantillon avec un inhibiteur de topoisomérase et/ou un inhibiteur de l'ADN gyrase ; c) traiter l'échantillon donné après les traitements aux étapes a) et b) avec un agent de coloration nucléaire ; et d) déceler un micro-organisme ou des micro-organismes traités avec l~agent colorant nucléaire dans l~échantillon par cytométrie de flux.
