Cancerous tumours vary significantly in their response to chemotherapy agents. Currently, it is difficult to reliably assess the level of tumour responsiveness to a chemotherapy regimen during or post-administration. Biomarkers of tumour sensitivity to chemotherapy agents have hitherto been unknown. Such a biomarker would expedite identification of nonresponsive patients, who may then switch to other, possibly more effective regimens. The present invention provides a method for determining tumour responsiveness to a chemotherapy agent, wherein RNA is isolated from tumour cells of a patient before, during, and after chemotherapy. The quality of the RNA can be determined by capillary electrophoresis and assignment of an RNA integrity number (RIN). RIN values during and/or after chemotherapy are inversely proportionate to the level of tumour responsiveness. The tumour RIN is an easily accessed biomarker of tumour responsiveness to chemotherapy. The tumour RIN may also be used to assess the efficacy of a chemotherapy regimen.
L'invention concerne des tumeurs cancéreuses qui varient significativement dans leur réponse à des agents chimiothérapeutiques. Actuellement, il est difficile d'évaluer fiablement le niveau de la réaction tumorale à un régime de chimiothérapie pendant l'administration ou post-administration. Des biomarqueurs de la sensibilité tumorale à des agents chimiothérapeutiques sont à ce jour inconnus. Un tel biomarqueur pourrait accélérer l'identification des patients insensibles, qui peuvent alors adopter d'autres régimes éventuellement plus efficaces. L'invention concerne un procédé permettant de déterminer la réaction tumorale à un agent chimiothérapeutique, dans laquelle ARN est isolé des cellules tumorales d'un patient avant, pendant et après la chimiothérapie. La qualité de l'ARN peut être déterminée par électrophorèse capillaire et affectation d'un numéro d'intégrité d'ARN (RIN). Les valeurs de RIN pendant et/ou après la chimiothérapie sont inversement proportionnelles au niveau de réaction tumorale. Le RIN tumoral est un biomarqueur facilement accessible de la réaction tumorale à la chimiothérapie. On peut également l'utiliser pour évaluer l'efficacité d'un régime chimiothérapeutique.
