The present invention relates to the expression of various forms of a novel receptor in hematopoietic and endothelial cells. A variant form of this receptor has been detected in brain cells and shown to bind to the obese gene product, leptin. In particular, the present invention relates to methods of using leptin to stimulate the growth and development of receptor-positive hematopoietic and endothelial cells in vitro and in vivo. In addition, this receptor is selectively expressed in hematopoietic progenitor cells with long- term repopulating potential. Thus, agents that specifically bind to this receptor may be used to identify and isolate progenitor cells for a variety of clinical applications.
La présente invention se rapporte à l'expression de différentes formes d'un nouveau récepteur dans les cellules hématopoïétiques et endothéliales. Une variante de ce récepteur a été détectée dans les cellules du cerveau et s'est avérée se lier au produit génique obèse, la leptine. Cette invention se rapporte notamment à des procédés d'utilisation de la leptine afin de stimuler la croissance et le développement des cellules positives hématopoïétiques et endothéliales du récepteur in vitro et in vivo. De plus, ce récepteur est exprimé de manière sélective dans les cellules souches hématopoïétiques présentant un potentiel de repopulation à long terme. Ainsi, les agents qui se lient spécifiquement à ce récepteur peuvent être utilisés pour identifier et isoler les cellules souches dans une grande variété d'applications cliniques.
