The invention provides methods and compositions for improved protein production. The method comprises the steps of: (a) introducing into a host cell a first nucleic acid sequence comprising a signal sequence operably linked to a desired protein sequence; (b) expressing the first nucleic acid sequence; (c) co-expressing a second nucleic acid sequence encoding a chaperone or foldase selected from the group consisting of bip1, ero1, pdi1, tig1, prp1,ppi1, ppi2, prp3, prp4, calnexin, and Ihs1; and (d) collecting the desired protein secreted from the host cell. The first nucleic acid sequence optionally comprises an enzyme sequence between the signal sequence and the desired protein sequence.

L'invention concerne des procédés et des compositions pour la production améliorée de protéines. Le procédé comprend les étapes consistant à : (a) introduire dans une cellule hôte une première séquence d'acides nucléiques comprenant une séquence de signal liée de manière opérationnelle à une séquence de protéines souhaitée; (b) exprimer la première séquence d'acides nucléiques; (c) co-exprimer une seconde séquence d'acides nucléiques codant une chaperone ou foldase sélectionnée dans le groupe comprenant bip1, ero1, pdi1, tig1, prp1, ppi1, ppi2, prp3, prp4, calnexine, et Ihs1; et (d) collecter la protéine souhaitée sécrétée par la cellule hôte. La première séquence d'acides nucléiques comprend facultativement une séquence d'enzymes entre la séquence de signal et la séquence de protéines souhaitée.
