A method of imaging a histological sample is provided which comprises stimulating autofluorescence in a sample using electromagnetic radiation of wavelength 750nm. Detection of the autofluorescence is undertaken by a CCD camera, typically with an excitation filter of 510nm disposed between the camera and the sample. The weak signal from autofluorescence is reinforced by manipulation of data acquired by the camera. Autofluorescence is detected from an upper face of the sample, an upper layer of the sample removed and a next face of the sample imaged. These steps are repeated to obtain information on autofluorescence throughout the sample. The method enables detection of features for virtually any thickness of slice, and in particular slices can range from 50µm to 1µm, with the method generally being used to analyse slices of thickness 5-1µm. There is also provided a method of preparing a histological sample for use in the above method, and apparatus (10) for imaging autofluorescence in histological samples.
L'invention concerne un procédé permettant de produire une image d'un échantillon histologique consistant à stimuler l'autofluorescence dans un échantillon au moyen d'un rayonnement électromagnétique d'une longueur d'onde de 750 nanomètres. La détection de l'autofluorescence s'effectue au moyen d'une caméra CCD, un filtre d'excitation de 510 nanomètres étant en général disposé entre la caméra et l'échantillon. Le signal faible provenant de l'autofluorescence est renforcé par la manipulation de données acquises par la caméra. L'autofluorescence est détectée à partir de la surface supérieure de l'échantillon, une couche supérieure de l'échantillon est retirée et on produit une image de la surface suivante de l'échantillon. Ces étapes sont répétées afin d'obtenir des informations sur l'autofluorescence dans tout l'échantillon. Ce procédé permet de détecter des caractéristiques pour presque toutes les épaisseurs de tranche, et en particulier des tranches allant de 50 microns à 1 micron, ce procédé étant généralement utilisé pour analyser des tranches d'une épaisseur de 5 à 1 micron. L'invention concerne également un procédé permettant de préparer un échantillon histologique à utiliser dans le procédé susmentionné, et un appareil (10) permettant de produire une image de la fluorescence dans des échantillons histologiques.
