The present invention relates to compounds of Formula (I) wherein X is -S- or - NH-; R1 is alkyl, alkenyl, arylalkyl, arylalkenyl or aryl-O-alkyl, wherein the aryl groups are optionally substituted by one or more substituents, selected from the group consisting of lower alkyl, lower alkoxy, halogen or lower halogen-alkyl; R2 is hydrogen, lower alkyl or cycloalkyl; R3/R4 may form together with the N-atom to which they are attached a non aromatic5, 6 or 7 membered ring, which may contain in addition to the N-atom one additional heteroatom selected from the group consisting of O, S or N, and wherein the ring is optionally substituted by hydroxy, lower alkyl, lower alkoxy, -NR2, - CONR2, -CO-lower alkyl or benzyl; or may form together with the N-atom to which they are attached a heterocyclic ring system, containing at least two rings and which may contain one or two additional heretoatoms, selected from the group consisting of N or O; R is hydrogen or lower alkyl; R5 is hydrogen or lower alkyl; and to pharmaceutically suitable acid addition salts thereof. It has been found that the compounds are active on the GABAB receptor and therefore useful for the treatment of anxiety, depression, epilepsy, schizophrenia, cognitive disorders, spasticity and skeletal muscle rigidity, spinal cord injury, multiple sclerosis, amyotrophic lateral sclerosis, cerebral palsy, neuropathic pain and craving associated with cocaine and nicotine, psychosis, panic disorder, posttraumatic stress disorders or gastro- intestinal disorders.
L'invention concerne des composés représentés par la formule (I) dans laquelle X représente -S- ou -NH-; R?1¿ représente un alkyle, un alcényle, un arylalkyle, un arylalcényle ou un aryl-O-alkyle, les groupes aryle étant éventuellement substitués par un ou plusieurs substituants, choisis dans le groupe constitué d'alkyle inférieur, d'alcoxy inférieur, d'halogène ou d'halogène-alkyle inférieur; R?2¿ représente un hydrogène, un alkyle inférieur ou un cycloalkyle; R?3¿/R?4¿ peuvent former ensemble avec l'atome d'azote auquel ils sont rattachés un noyau non aromatique à 5, 6 ou 7 éléments, qui peut contenir outre l'atome d'azote un hétéroatome supplémentaire choisi dans le groupe constitué de O, S ou N, et ce noyau pouvant éventuellement substitué par un hydroxy, un alkyle inférieur, un alcoxy inférieur, -NR¿2?, -CONR¿2?, -CO- alkyle ou benzyle inférieur; ou peuvent former ensemble avec l'atome d'azote auquel ils sont rattachés un système de noyau hétérocyclique, ce système contenant au moins deux noyaux et pouvant contenir un ou plusieurs hétéroarômes supplémentaires choisis dans le groupe constitué de N ou O: R représente un hydrogène ou un alkyle inférieur; R?5¿ représente un hydrogène ou un alkyle inférieur. L'invention concerne également des sels d'addition acide de ces composés acceptables sur le plan pharmaceutique. On a découvert que ces composés sont actifs sur le récepteur GABA<SB>B</SB> et par là même utiles dans le traitement de l'anxiété, de la dépression, de l'épilepsie, de la schizophrénie, de troubles cognitifs, de la spasticité et de la rigidité du muscle squelettique, du traumatisme médullaire, de la sclérose en plaques, de la sclérose latérale amyotrophique, de l'infirmité motrice cérébrale, de douleurs neuropathiques et de l'état de manque associé à la cocaïne et à la nicotine, de la psychose, du trouble panique, des troubles de stress post-traumatiques ou des troubles gastro-intestinaux.
