The present invention relates generally to the field of tissue modification, and in particular, but not exclusively, to methods of modifying cells of ocular and particularly corneal tissue to produce immunoglobulins or immunoglobulin fragments. The invention also relates to modified ocular cells and tissues, to expression vectors utilised in such methods, to methods of xeno- and allo-transplantion using the modified tissue and to methods of therapy of ocular diseases and disorders. In a preferred embodiment of the invention there is provided a method of modifying cells of ocular tissue to produce an immunoglobulin or immunoglobulin fragment of interest, which comprises exposing ocular tissue to an effective concentration for transfection of an expression vector comprising a nucleotide sequence encoding for the immunoglobulin or immunoglobulin fragment, for a period sufficient to allow transfection, such that cells of said ocular tissue produce the immunoglobulin or immunoglobulin fragment.

L'invention concerne en général le domaine de la modification de tissus, et en particulier, mais pas exclusivement, des procédés de modification de cellules de tissus oculaires et en particulier de cellules de tissus cornéens afin d'obtenir des immunoglobulines ou des fragments d'immunoglobuline. L'invention concerne également des cellules et des tissus oculaires modifiés, des vecteurs d'expression utilisés dans lesdits procédés, des procédés de xénogreffe et d'allogreffe faisant intervenir lesdits tissus modifiés et des procédés de traitement de troubles et de maladies oculaires. Dans un mode de réalisation préféré de l'invention, un procédé de modification de cellules de tissus oculaires permettant de produire une immunoglobuline ou un fragment d'immunoglobuline d'intérêt consiste à exposer des tissus oculaires à une concentration efficace, en vue d'une transfection, d'un vecteur d'expression qui comprend une séquence nucléotidique codant l'immunoglobuline ou le fragment d'immunoglobuline, pendant une durée suffisante permettant d'assurer la transfection, de façon que les cellules desdits tissus oculaires produisent ladite immunoglobuline ou ledit fragment d'immunoglobuline.
